Differentation between Fusarium Fujikuroi and Fusarium Proliferatum based on mating studies and phlogenetic studies

Fusarium proliferatum is most likely to be confused with F. fujikuroi. Morphologically F. proliferatum and F. fujikuroi can not be effectively differentiated solely on morphology and are sibling species. PDA (potato dextrose agar) culture for both fusarium form white mycelia that become purple with age. The morphology and formation of macroconidia and microconidia are also indistinguishable. F.proliferatum forms slender, thin-walled, relatively straight macroconidia and typical of those produced by species in the F. fujikuroi. The microconidia of F.fujikuroi and F.proliferatum are oval or club shaped with a flattened base and 0- to 1-septate. A few pyriform microconidia may be present in some isolates for both species.(reff:lab manual)

Since F.Fujikuroi is effectively morphologically indistinguishable from F.Proliferatum so they can be accurately distinguish by using mating tests or by DNA sequences. The separation between them as biological species is not complete (1236), and naturally occurring hybrids between these two species have been recovered (1234). Phylogenetically, F. proliferatum and F. fujikuroi are very closely related as well (1596, 1690), although their karyotypes are different (2374).

Nowdays, molecular techniques to identify and group species are becoming important. Molecular method was easier and cheaper. The answers are less ambigous and different people will make same observation.The sort of method to use are genotyping and DNA sequences. The processs involving DNA extraction, PCR amplification using primer, cycling for sequence using primer with different concentration, sequencing and lastly homology search with Fusarium database.BLA BLA BLA


Since the differention between F.fujikuroi and F.proliferatum seem to be still unclear, the objective of this study is to identify species by amplification and DNA sequencing and also by their mating studies.
